By car and by train.
You can park your car directly in predisposed pay-gareges at Piazzale Roma, the only point reached by car in Venice, or the island of Tronchetto connected to Venice by a funicular called People Mover 3 mins far from Piazzale Roma. The main train station in Venice is Stazione F.S. Venezia S. Lucia, on the Grand Canal in the northwest corner of the city. Please note that some trains do not terminate at S. Lucia, stopping only at Venezia-Mestre station on the mainland. However, there are frequent connections to Venezia S. Lucia leaving from platform n° 1.
By plane.
Getting to Venice by plane is very convenient, 2 are the nearest airports, in Tessera Marco Polo or Treviso Antonio Canova.
